چکیده

We designed bioinspired cross-linkers based on desmosine, the cross-linker in natural elastin, to prepare hydrogels with thiolated hyaluronic acid. These short, rigid cross-linkers are based on pyridinium salts (as in desmosine) and can connect two polymer backbones. Generally, the obtained semi-synthetic hydrogels are form-stable, can withstand repeated stress, have a large linear-elastic range, and show strain stiffening behavior typical for biopolymer networks. In addition, it is possible to introduce a positive charge to the core of the cross-linker without affecting the gelation efficiency, or consequently the network connectivity. However, the mechanical properties strongly depend on the charge of the cross-linker. The properties of the presented hydrogels can thus be tuned in a range important for engineering of soft tissues by controlling the cross-linking density and the charge of the cross-linker.

Charged Triazole Cross-Linkers for Hyaluronan-Based Hybrid Hydrogels

Polyelectrolyte hydrogels play an important role in tissue engineering and can be produced from natural polymers, such as the glycosaminoglycan hyaluronan. In order to control charge density and mechanical properties of hyaluronan-based hydrogels, we developed cross-linkers with a neutral or positively charged triazole core with different lengths of spacer arms and two terminal maleimide groups...

Extremely stretchable thermosensitive hydrogels by introducing slide-ring polyrotaxane cross-linkers and ionic groups into the polymer network

Stimuli-sensitive hydrogels changing their volumes and shapes in response to various stimulations have potential applications in multiple fields. However, these hydrogels have not yet been commercialized due to some problems that need to be overcome. One of the most significant problems is that conventional stimuli-sensitive hydrogels are usually brittle. Here we prepare extremely stretchable t...

General. Linkers used in preparation of modified hyaluronan (HA) derivatives were synthesized according to stated literature procedures. Their structures are given in Scheme S1. Particularly, 3,3'dithiobis(propionic hydrazide) 11 and 2-(2-pyridinyldithio)ethyl hydrazinecarboxylate 32 were used to attach thioland 2-dithiopyridyl-terminated side chains to HA backbone respectively. 3-(Acrylamido-1...

Hyaluronan microspheres for sustained gene delivery and site-specific targeting.

Hyaluronan is a naturally occurring polymer that has enjoyed wide successes in biomedical and cosmetic applications as coatings, matrices, and hydrogels. For controlled delivery applications, formulating native hyaluronan into microspheres could be advantageous but has been difficult to process unless organic solvents are used or hyaluronan has been modified by etherification. Therefore, we pre...

Mechanically strong triple network hydrogels based on hyaluronan and poly(N,N-dimethylacrylamide).

Hyaluronan (HA) is a natural polyelectrolyte with distinctive biological functions. Cross-linking of HA to generate less degradable hydrogels for use in biomedical applications has attracted interest over many years. One limitation of HA hydrogels is that they are very brittle and/or easily dissolve in physiological environments, which limit their use in load-bearing applications. Herein, we de...
